Abstract

|
Mutation is the 69bp region of rpoB gene, encoding¥âsubunit of DNA depedent RNA polymerase, is considered as a principal molecular mechanism of rifampicin(RFP) resistance of Mycobacterium tuberculosis. Line Probe Assay(LiPA) was developed
recently,
whose principle is hybridization of PCR fragment amplified with clinical isolate on the strip previously attached 69bp region frequently occurring mutation. 10 RFP susceptible and 36 RFP resistant strains(MIC¡Ã40¥ìg/ml in the Lowenstein-Jensen
medum)
were selected for LiPA. As a results of LiPA and sequencing, 10 of RFP susceptible strains showed identical pattern with M. tuberculosis H37Rv on LiPA. 34(94.4%) of 36 strains were mutated within 69bp region and two were identical with M.
tuberculosis
H37Rv. Therefore LiPA could be a useful tool to discriminate at least RFP resistant strain in clinical isolates.
